Initially earning a Bachelor of Arts and a Master of Arts from Boston University, Dr. D'Agostino later graduated with a Doctor of Philosophy at Harvard University in 1968. He began his career teaching mathematics at Boston University in 1964, advancing to the posts of an assistant professor in 1964, an associate professor in 1971 and a professor in 1976. In 1976, he was appointed as the associate dean of the Graduate School at Boston University, and he expanded his focus to include course offerings in public health, law, biostatistics and epidemiology through the 1980s. Dr. D'Agostino continues to teach at Boston University and Tufts University, for which he was named the director of graduate programs in biostatistics.

Currently consulting for dozens of high-profile organizations, including the United States Food and Drug Administration and international pharmaceutical manufacturers including Millenium, Bayer, Sage Therapeutics and Sanofi, Dr. D'Agostino was the lead in a joint study by the American College of Cardiology and the American Heart Association to estimate heart disease risk factors. He further flourished as a principal investigator in the Framingham Heart Study and a special scientist on projects with Tufts Medical Center and the National Institutes of Health. Likewise, Dr. D'Agostino has contributed to numerous pharmaceutical and clinical studies, grant and policy proposals and panels.

Dr. D'Agostino has been active as a lecturer for Cambridge Heart, the American Public Health Association, the University of Florida, the University Hospital of Geneva and the University of Toledo Medical School, among many others. He also serves as an editor of Statistics in Medicine and the New England Journal of Medicine. Throughout his career, Dr. D'Agostino has held many leadership and administrative positions in the American Heart Association, the American Statistical Association and the American Public Health Association.

Dr. D'Agostino is the author of several texts on clinical research and specialty mathematics for healthcare professions, including "Pharmaceutical Studies Using SAS" in 2008 and "Therapeutic Strategies in Cardiovascular Risk" in 2010. He has provided his wealth of knowledge to more than 800 articles published in professional journals. Participating on the editorial boards of dozens of publications, Dr. D'Agostino is a reviewer for major publishers including Houghton-Mifflin, Duxbury Press and Prentice Hall.

Dr. D'Agostino has been recognized with more than a dozen prestigious awards, fellowships and accolades. He notably accepted Achievement and Distinguished Service Awards from the American Public Health Association and twice a Special Citation from the Commissioner of the United States Food and Drug Administration. Moreover, Dr. D'Agostino was presented with Metcalf Award for Teaching from Boston University.
