SHIRLEY, NY, May 29, 2015 /24-7PressRelease/ -- Researchers from the Creative Animodel Institute use CRISPR/Cas genetic regulatory system, manipulating mouse genome with one step into the reporter gene and the conditional allele. Now we just need several weeks rather than a few years time to generate such a complex engineering allele animals and also to construct models of disease or to research gene function using these animals.
Creative Animodel Institute said: "We have previously used CRISPR/Cas to mutate gene, but it can not predict the characteristics of these targeted mutations. Now we can define the specificity deletion by two times cutting. We can generate conditional mice one step, easily and very effectively insert DNA fragments of up to 3000 base pairs. In the past, however, we need much more work to to carry building such a mouse."
CRISPR/Cas system (clustered regularly interspaced short palindromic repeat/CRISPR associated) is a natural immune system found in most bacteria and archaea which can be used to against viruses and other pathogens of bacterial invasion. Scientists recently alter the genome of mice and human cells quickly and effectively by using the defense mechanism. However, until now, researchers start to utilize CRISPR/Cas to construct the most useful tool of genetic study: conditional mutant mice.
A conditional mutation mouse contains one or a group of genes which can make use of a specific signal to turn on or off. By opening or closing these genes, scientists can tease out the function of some genes in biological function and disease. Previously, scientists required embryonic stem to construct such a model organism as well as underwent a complex and time-consuming process. Unfortunately, scientists could only control the EScs of mice and rats, limiting the study. Using CRISPR/Cas, Creative Animodel lab constructed mice with a conditional allele and a plurality of marker genes to report whether the allele is expressed normally.
Besides, the experiment had reduced the fear of CRISPR/Cas miss activity. Creative Animodel said: "recently, several studies about human cancer cell lines presented the concerns on the specificity of CRISPR/Cas. Our research shows that nonspecific DNA cutting may occur, but they are rare and can predict."
The latest research in Creative Animodel institution opens up new avenues for future research. "In this study, we describe those methods that will greatly speed up the process to construct transgenic modified animals. We want to construct a model of complex diseases by using this approach with CRISPR/Cas," said the researchers.
Since CRISPR/Cas does not depend on ESCs, it can be used to do genetic transformation on any animals including domestic animals.
We haven't tried, but we want to control genome project in large animals through transforming the CRISPR/Cas system.
